Cryopreservation of sperm

The ability to have children in the future does not always depend on desire alone. A man's fertility is affected by age, illness, treatment of serious pathologies, and life circumstances. Sperm cryopreservation allows you to preserve sperm in a functional state for years for use in the future when it is needed, for example, before chemotherapy, surgery, participation in IVF programs or postponing parenthood.

Modern methods of sperm freezing ensure high survival of germ cells after thawing, making this approach an effective way to preserve reproductive potential and chances of successful conception in the future. Next, let's take a closer look at the features of sperm cryopreservation, when it is really necessary, and how the procedure works.

What is sperm cryopreservation?

Sperm cryopreservation is a method of long-term preservation of male germ cells by freezing them at ultra-low temperatures using liquid nitrogen. The biomaterial is gradually cooled to -196 °C. At this temperature, all metabolic processes in the cells virtually stop, which allows sperm to be preserved for a long time with minimal loss of fertilization ability.

Before freezing, special protective substances are added to the biomaterial to prevent damage to the cells during cooling and subsequent storage. The procedure is performed in specialized laboratory conditions using appropriate equipment.

The sperm is placed in sealed sterile cryotubes or special small-volume containers, and then placed in Dewar vessels. These are vacuum-insulated tanks designed to safely store liquid nitrogen and maintain a consistently low temperature. This system guarantees reliable protection of biological material from temperature fluctuations and allows it to be stored for decades without losing its functional properties.

It's interesting to know! The first attempts to preserve sperm at low temperatures were made by the Italian scientist Lazzaro Spallanzani back in 1776. He proved that after cooling, sperm can resume their activity.

But the real breakthrough came in 1949, when researcher Christopher Poldge discovered the protective effect of special substances that allowed sperm to be safely frozen and stored for a long time. This discovery became the basis of modern reproductive medicine and made it possible to give birth to healthy children using assisted reproductive technologies.

Indications for freezing sperm

Cryopreservation is an effective way to preserve a man's reproductive potential in cases where there is a risk of sperm quality deterioration or parenthood is planned in the future. The procedure allows you to record the current state of sperm and use them for in vitro fertilization when it is necessary.

The main indications for sperm freezing:

  • preparation for chemotherapy or radiation therapy, which can damage sperm;
  • if you are planning to have surgery on the testicles, prostate gland or other organs of the reproductive system;
  • a decrease in sperm count or quality, which can progress over time;
  • desire to postpone parenthood for personal, medical or professional reasons;
  • working in harmful conditions (exposure to toxins, high temperatures, radiation, chemicals);
  • Preparing for assisted reproductive technology programs to have a stock of biomaterial;
  • difficulties with obtaining sperm on the day of fertilization (psychological or physiological reasons);
  • planning a vasectomy (surgical sterilization) with the possibility of preserving the chance of procreation;
  • service in the army or other circumstances involving a risk to health and life;
  • hormonal treatment or medical conditions that may affect fertility.

Sperm freezing makes sure that the possibility of becoming a father will remain even in the presence of unfavorable factors.

Advantages of the procedure

Sperm cryopreservation is a special method of preserving male biological material, i.e. sperm. Sperm is frozen by the method of instant freezing - vitrification.

Unlike the slow freezing method, vitrification does not form ice crystals in the sperm , so the natural structure of sperm is fully preserved. For freezing, a special cryoprotective substance is used, which turns into a glassy state under the influence of low temperatures.

How the procedure takes place at the IVMED clinic

Sperm cryopreservation is a simple, safe and non-invasive procedure that usually takes about 20-30 minutes. Upon receipt of the biomaterial, the sample is immediately labeled with an individual code, which eliminates the risk of confusion. All information about IVMED patients is entered into a secure electronic database in complete confidentiality. This ensures accurate identification of the material and its safe use in the future.

Ejaculate donation in our clinic takes place in a comfortable, specially prepared room. The material is obtained by masturbation in a sterile container, after which it is immediately transferred to laboratory specialists for quality assessment and further preparation for freezing. All stages are carried out in compliance with medical standards, and the sample is labeled with an individual code to ensure safety and accurate identification.

Preparation for ejaculate donation

Before cryopreservation of sperm, a man is usually recommended to undergo basic examinations to assess the condition of the reproductive system:

  • spermogram - to assess the concentration, motility, and morphology of spermatozoa;
  • testing for sexually transmitted infections (HIV, hepatitis B and C, syphilis, etc.);
  • general blood and urine tests - to assess the general state of health;
  • hormonal tests - if indicated, to assess the level of testosterone and other sex hormones;
  • biochemical blood tests - if necessary, to exclude contraindications to the procedure.

Proper preparation for ejaculate donation is equally important. It helps to obtain the highest quality biomaterial, which is especially important for its successful freezing and further use. Basic rules:

  1. Abstain from ejaculation for 2-5 days. This is the optimal period for the accumulation of a sufficient number of active sperm.
  2. Exclusion of alcohol 48-72 hours before the procedure, as it worsens the quality of sperm.
  3. Avoid overheating (saunas, hot baths, tanning beds, overheating in the bath or at work) for 3-5 days.
  4. Perform normal hygiene procedures before handing over the material.
  5. Avoid severe stress, lack of sleep, and physical exhaustion the day before.

In case of fever or acute illnesses, the procedure should be postponed, and at least 1-2 weeks should pass after recovery.

You should also inform your doctor about taking any medications, especially antibiotics or hormones. Following these recommendations helps to obtain the best quality sample for further cryopreservation and successful preservation of reproductive potential.

The process of freezing and storage in a cryobank

After receiving the ejaculate, the sample undergoes laboratory processing to maximize the viability of sperm during freezing. Special protective substances, cryoprotectants, are added to the biomaterial to prevent cell damage when exposed to ultra-low temperatures.

The process of freezing and storing sperm involves several stages:

  1. Laboratory evaluation of sperm. The concentration, motility and viability of spermatozoa are determined.
  2. Adding cryoprotectants. Special media protect cells from the formation of ice crystals.
  3. Gradual cooling. The sample is cooled in stages to adapt the cells to low temperatures.
  4. Freezing in liquid nitrogen. Sperm is placed in special sterile cryocontainers and immersed in liquid nitrogen tanks at a temperature of about -196 °C.
  5. Long-term storage of sperm in a cryobank. Samples are stored in special containers with constant temperature control and security.

In this state, sperm can be stored for years without losing their properties, and this period is not limited. If necessary, the material is thawed and used in assisted reproductive technology programs, in particular for fertilization of an egg.

Modern cryotechnologies provide high efficiency of sperm preservation and give a chance for successful parenthood at any age.

Use of sperm after cryopreservation in ART programs

Freezing sperm makes it possible to use it at any time. Controlled thawing is performed to gradually restore the temperature of the biomaterial and preserve the functional properties of the cells.

After thawing, sperm can be used in various programs of assisted reproductive technologies:

  • IVF - fertilization of an egg in the laboratory with the subsequent transfer of embryos to the uterine cavity;
  • ICSI - the doctor selects one viable sperm and injects it directly into the egg;
  • intrauterine insemination - the prepared material is inserted into the uterine cavity for natural fertilization.

Our clinic offers the use of cryomaterial as part of a comprehensive infertility treatment. Also, sperm freezing is the best choice for those who want to postpone parenthood but maintain high chances of successful conception at the most favorable time.
